2. Indications
It blocks orexin receptors, regulates the sleep-wake cycle, and helps improve insomnia (such as difficulty falling asleep and sleep maintenance disorders).
3. Mechanism of action
Vonoresen is an orexin 1 (OX1) and orexin 2 (OX2) receptor antagonist, with binding affinity (Ki value) of 0.460 nmol/L and 0.374 nmol/L for human OX1 and OX2 receptors, respectively. It induces the transition from wakefulness to sleep by inhibiting the binding of the wakefulness-promoting neuropeptides orexin A and B to OX1 and OX2 receptors.
4. Usage and Dosage
Usual dosage: Adults usually take 5 mg once a day, orally before bedtime.
Dosage adjustment: The dosage may be increased or decreased as needed based on symptoms, but the daily dose should not exceed 10 mg.
5. Precautions
Use with caution in patients with narcolepsy or cataplexy, organic brain disorders, or respiratory dysfunction. Patients with mild to moderate hepatic impairment require close monitoring after administration; contraindicated in patients with severe hepatic impairment. Pregnant women should only use this medication when the benefits outweigh the risks; breastfeeding women should weigh the benefits and risks before deciding whether to breastfeed. Contraindicated in children. Dosage adjustments should be made individually for elderly patients.
