2. Indications
Blocks orexin receptors, regulates the sleep-wake cycle, and helps improve insomnia (such as difficulty falling asleep and maintaining sleep).
3. Mechanism of Action
Vonorexant is an antagonist of orexin 1 (OX1) and orexin 2 (OX2) receptors, with binding affinities (Ki values) of 0.460 nmol/L and 0.374 nmol/L for human OX1 and OX2 receptors, respectively. It induces the transition from wakefulness to sleep by inhibiting the binding of the wakefulness-promoting neuropeptides orexin A and B to OX1 and OX2 receptors.
4. Dosage and Administration
Conventional dosage: Adults usually take 5mg once daily, orally, immediately before bedtime.
Dosage adjustment: The dose can be adjusted appropriately according to symptoms, but the daily dose should not exceed 10mg.
5. Precautions
Use with caution in patients with narcolepsy or cataplexy, organic brain disorders, or respiratory dysfunction. Close monitoring is required after administration to patients with mild to moderate hepatic impairment; contraindicated in severe hepatic impairment. Pregnant women should only use if the benefits outweigh the risks. For lactating women, the benefits and risks of breastfeeding should be weighed. Contraindicated in children. Dosage should be adjusted individually for elderly patients.
